Poundstone started performing stand-up in Boston in 1979 and traveled the country by bus, honing her craft in comedy clubs. She settled for a time in San Francisco, where her improvisational style gained attention, before moving to Los Angeles with encouragement from peers including Robin Williams.

She gained national exposure through television appearances and HBO specials. Her 1990 special Cats, Cops and Stuff earned her a CableACE Award for Best Stand-Up Comedy Special—the first time a woman won in that category.
